4 cases reported with biallelic variants (3 chets & 1 hom) and variable phenotypes that included no to profound developmental delay (3 with dev delay), all with abnormal transferrin glycosylation. Also, an in vitro functional assay that supports impaired function for the 2 (both frameshift variants) of the 5 variants that have been reported in the 3 cases. The authors suggested the assay could not differentiate hypomorphs from benign variants. Assigned MODERATE by ClinGen CDG GCEP in 2023.
