Two clinical forms of methemoglobinemia due to deficiency of methemoglobin reductase: type 1 (deficiency of b5R is isolated to erythrocytes) and type 2 (deficiency of b5R in all cell types) (MIM: 250800).
Type 2 presents with neurological damage manifested by severe psychomotor retardation, hypotonia, microcephaly and neuroimaging abnormalities. Type 2 usually caused by full stops or deletions located in consensus FAD or NADH-binding sites of enzyme causing loss of protein expression or enzymatically inactivate proteins leading to severe neurological impairment (PMID: 38303731).
