Features reported in Amish population, related individuals, with frameshift founder variant - c.394insA (PMID: 20170897)- organomegaly, failure to thrive, developmental delay, dysmorphic features, and autoimmune inflammatory cell infiltration of the lungs, liver, and gut. Growth stunted. Phenotypic overlap in 2nd unrelated reported case from (PMID: 31091003) compound het frameshift variants: c.726del and c.2125_2128del .
There is supportive functional data and animal studies (mice).
