Biallelic variants in MMACHC cause methylmalonic aciduria and homocystinuria type cblC. Neuropathy (particularly peripheral neuropathy (sensorimotor polyneuropathy)) is a presenting feature in affected individuals, accompanied by cognitive impairment, psychiatric symptoms, renal dysfunction and gait disturbance.
