Variants have been reported in >3 affected probands however the variants are very common (FAF >0.1%) in gnomAD v4.1 for AD condition.
Supportive functional depicting LoF mechanism and animal models recapitulating human phenotype was conducted.
ClinGen requires further information to upgrade Limited classification.
Further evidence is required to determine whether there are common disease causing variants in this gene, therefore this gene-disease association is downgraded to Amber.
