9 families with heterozygous variants causing severe early‑onset obesity, often with hyperphagia and neuro‑behavioral features) reported across multiple papers. Variants are generally missense with little supporting data. The PMID 32493978 report provides the first experimental functional validation (cell‑based assays showing impaired pro‑BDNF processing and loss‑of‑function).
